Description
Alpha-1 antitrypsin (AAT) is a potent, nonspecific inhibitor of proteolytic enzymes.
It is produced in the liver and by macrophages. It is an acute-phase protein whose concentration increases four- to five-fold during the acute phase of inflammation. In plasma, it inactivates leukocyte elastase released by granulocytes during the acute phase of inflammation and also binds trypsin, chymotrypsin, collagenase, plasmin, and thrombin.
Congenital deficiency of this protein is caused by a defect in AAT secretion from liver cells, which results in the accumulation of protein deposits in hepatocytes, leading to liver cirrhosis. AAT deficiency in plasma is a risk factor for the development of pulmonary emphysema and bronchiectasis. Low AAT concentrations are also observed in protein-losing syndromes and neonatal respiratory distress syndrome.
