Description

The U1-snRNP complex is a nuclear ribonucleoprotein particle composed of uridine-rich RNA, a set of proteins known as the 70-kDa U1-RNP, and an Sm (Smith antigen) molecule.
Antibodies to the snRNP complex belong to a heterogeneous group of antinuclear antibodies associated with autoimmune diseases. Antibodies to the snRNP complex react specifically with both the Sm antigen and a set of proteins known as the 70-kDa U1-RNP. They are typically present in patients with systemic lupus erythematosus (SLE) and mixed connective tissue disease (MCTD), but also occur in Sjögren's syndrome, scleroderma, and polymyositis.
Differentiation of autoantibodies by ELISA using a specific antigen is helpful in differentiating SLE from MCTD. Antibodies against the U1-RNP 70kDa protein complex are present in approximately 95% of patients with MCTD and in 40% of patients with SLE (the detected presence of anti-U1 RNP antibodies is a typical symptom of Sharp's syndrome). Antibodies against Sm are highly specific for SLE.
