Antiphospholipid syndrome (APS), also known as Hughes syndrome, is characterized by symptoms of vascular thrombosis and recurrent miscarriages, along with the presence of serum antibodies specific for anionic phospholipids and phospholipid-binding proteins. The clinical criteria for antiphospholipid syndrome include:
1. vascular thrombosis;
2. spontaneous miscarriages.
The following hypotheses regarding the pathogenetic role of antiphospholipid antibodies have been proposed: antiphospholipid
antibodies inhibit the modulating effect of thrombin on endothelial cells, which may impair prostacyclin release and inhibit protein C activation; antiphospholipid antibodies recognize the antiphospholipid/phospholipid anion complex on the surface of endothelial cells, which induces prothrombotic reactions involving prothrombin; and antiphospholipid antibodies may increase the affinity of prothrombin for phospholipids and thus induce thrombotic mechanisms.
